EK protease is a common tool enzyme used to excise affinity tags on fusion
proteins. It has strong site specificity and strictly recognizes the five-amino
acid sequence DDDDK↓. The cleavage
site is between aspartic acid and lysine. between. The commonly used five amino
acid sequence is: Asp-Asp-Asp-Lys↓.
The
recombinant EK protease can achieve the best activity at pH 7.4 and 23 °C, but
it is active in a wide range of pH 4.5 to 9.5 and temperature of 4 to 45 °C, so
that the choice of reaction conditions can be based on the target protein. Be
flexible. In addition, the His-tag contained in rEK can be used to remove it by
Ni-NTA resin to achieve the purpose of purifying unlabeled protein.
